How to make Nicotine.

Nicotine extraction from tobacco leaves
(1) Weigh dry tobacco leaves
(2) Macerate tobacco leaves in methanol
(3) Acidify with sulfuric acid (H2SO4) 2M to pH = 3
(4) Evaporate alcohol, using a water bath, to a small volume
(5) Transfer this to an extraction vessel
(6) Add 60 ml of H2SO4 2M and 50 ml of diethyl ether
(7) Shake carefully
(8) Allow layers to separate
(9) Discard ether layer; the nicotine is dissolved in the aqueous layer (1st extraction) (Determine which layer is which, by the drop test)
(10) Extract aqueous layer with ether three more times (3 X 50 ml), discarding the ether layer each time
(11) Cool the aqueous layer
(12) Basify with ammonium hydroxide to pH = 10
(13) Extract again with ether (8 X 25 ml), this time keeping the ether layer (2nd extraction) (Add ether to aqueous solution, remove ether and keep it, add new ether to aqueous solution, remove ether and keep it…)
(14) Pool ether extracts
(15) Extract ether solution with HCl 0.5M (4 X 50 ml) and discard ether layer (3rd extraction)
(16) Basify the aqueous solution with NaOH 10% to pH = 11
(17) Extract aqueous solution with chloroform (7 X 50 ml) (4th extraction)
(18) Using anhydrous sodium sulfate, the chloroform extracts are dried, filtered and evaporated in a fume cupboard.
